Duke Nukem certainly has more popularity as a character and a bit more relatability as an Ash (Evil Dead) archetype, but Serious Sam (especially the engine) has a far superior design, feature set, gameplay flow, and especially cooperative emphasis. The action is more fast-paced, the monsters/weaponry is WAAY over-the-top, and the "puzzles" and story construction are so extremely satirical that it's almost a parody in itself.
Also if you want to get down to the nitty gritty, Duke is a copy of MANY other existing characters -- Ash, like I mentioned, Arnold, Van Damme, and a slew of other older action-movie stars. Sam is NOT a copy of these (nor a copy of Duke), but he IS trying to mimic (poorly) the same concepts in a mocking manner.
