You can use practically any graphics program to make sprites: Photoshop, Paint Shop Pro, PainterX, Paint, Paint.NET, Gimp, etc.

There's a large number of formats which end in the extension "spr", so some more information as to what you want to use it for would be very helpful, otherwise it would be next to improbable to point you in the right direction without either immense luck with guess work or psychic abilities.

Alex is right. Sprite is a generic term coined to represent a 2D image (usually with some level of transparency). There's no default format for a sprite as it can be represented in dozens of image types. Any .spr extension representing a sprite would be a proprietary file type for a specific application or usage.
